Book excerpt: From the author of An Eccentric Engagement comes a Regency romance celebrating the witty and romantic world that fans of Georgette Heyer have fallen in love with. “Simpson excels at imbuing her realistic characters with subtle depths!” —John Charles, American Library Association As chaperone to the Season’s most desirable young miss, Amy Corbett has two specific duties: to ensure that her charge’s reputation remain spotless, and to see her betrothed to an exceptional beau. Given that the coquettish Lady Rowena manages to maintain an air of propriety while being a flirt of the highest caliber, Amy knows her job should be easy—if only the calculating young aristocrat weren’t so set on toying with men’s hearts, including that of her latest prey, the debauched Lord Dante Pierson. Like other gentlemen of high standing and low tastes, Lord Pierson’s restless desire for excitement had driven him into a life of squalor and scandalous social blunders. Finding himself drunk and mud-spattered in a gutter one night, he spies an angelic apparition in a passing carriage and is overcome by a will to reform—surely such a woman could give him hope of rebirth. When he learns that the anonymous beauty is Lady Rowena, he vows to coax that flicker of warmth into a flame. As Rowena lures the unsuspecting Pierson ever closer to heartbreak, Amy realizes that for all the talk of his wastrel ways, he is actually a gentle and caring man to be championed and cherished. And in an unguarded moment, the unassuming Amy and downtrodden Lord Pierson will discover that they possess kindred souls—and will make a daring decision to follow their own true path to happiness and love . . . This book was originally published as Lord Pierson Reforms.

Available in PDF, EPUB and Kindle. Book excerpt: Members of the HSP70 family form a central hub of the molecular chaperone network, controlling protein homeostasis in prokaryotes and in the ATP-containing compartments of the eukaryotic cells. The heat-inducible form HSPA1A (HSP70), its constitutive cytosolic cognate HSPA8 (Hsc70), its endoplasmic reticulum form HSPA5 (BiP), and its mitochondrial form HSPA9 (Mortalin), as well as the more distantly related HSPHs (HSP110s), make up 1-2 % of the total mass of proteins in human cells. They use the energy of ATP-hydrolysis to prevent and forcefully revert the process of protein misfolding and aggregation during and following various stresses, presumably by working as unfoldases to lift aberrant conformers out of kinetic traps. As such, HSP70s, in cooperation with their J-domain co-chaperones and nucleotide exchange factors (NEFs) and co-disaggregases, form an efficient network of cellular defenses against the accumulation of cytotoxic misfolded protein conformers, which may cause degenerative diseases such as Parkinson's and Alzheimer's disease, diabetes, and aging in general. In addition to their function in repair of stress-induced damage, HSP70s fulfill many housekeeping functions, including assisting the de novo folding and maturation of proteins, driving the translocation of protein precursors across narrow membrane pores into organelles, and by controlling the oligomeric state of key regulator protein complexes involved in signal transduction and vesicular trafficking. For reasons not well understood, HSP70s are also found on the surface of some animal cells, in particular cancer cells where they may serve as specific targets for cancer immunotherapy. Here, we gathered seven mini reviews, each presenting a complementary aspect of HSP70’s structure and function in bacteria and eukaryotes, under physiological and stressful conditions. These articles highlight how, the various members of this conserved family of molecular chaperones, assisted by their various J-domain and NEF cochaperones and co-disaggregases, harness ATP hydrolysis to perform a great diversity of life-sustaining cellular functions using a similar molecular mechanism.

This book was released on 2020-03-16 with total page 440 pages. Available in PDF, EPUB and Kindle. Book excerpt: Replicating and Repairing the Genome provides a concise overview of the fields of DNA replication and repair. The book is particularly appropriate for graduate students and advanced undergraduates, and scientists entering the field or working in related fields. The breadth of information regarding DNA replication and repair is vast and often difficult to absorb, with terminology that differs between experimental systems and with complex interconnections of these processes with other cellular pathways. This book provides simple conceptual descriptions of replication and repair pathways using mostly generic protein names, laying out the logic for how the pathways function and highlighting fascinating aspects of the underlying biochemical mechanisms and biology. The book incorporates extensive and informative diagrams and figures, as well as descriptions of a number of carefully chosen experiments that had major influences in the field. The process of DNA replication is explained progressively by starting with the system of a simple bacterial virus that uses only a few proteins, followed by the well-understood bacterial (E coli) system, and then culminating with the more complex eukaryotic systems. In the second half of the book, individual chapters cover key areas of DNA repair — postreplication repair of mismatches and incorporated ribonucleotides, direct damage reversal, excision repair, and DNA break repair, as well as the related areas of DNA damage tolerance (including translesion DNA polymerases) and DNA damage responses. The book closes with chapters that describe the huge impact of DNA replication and repair on aspects of human health and on modern biotechnology.

Available in PDF, EPUB and Kindle. Book excerpt: The term “heavy metals” is used as a group name of toxic metals and metalloids (semimetals) causing contaminations and ecotoxicity. In strict chemical sense the density of heavy metals is higher than 5 g/cm3. From biological point of view as microelements they can be divided into two major groups. a. For their physiological function organisms and cells require essential microelements such as iron, chromium (III), cobalt, copper, manganese, molidenium, zinc. b. The other group of heavy metals is toxic to the health or environment. Of highest concern are the emissions of As, Cd, Co, Cu, Hg, Mn, Ni, Pb, Sn, Tl. The toxicity of heavy metals is well known at organizational level, while less attention has been paid to their cellular effects. This book describes the toxicity of heavy metals on microorganisms, yeast, plant and animal cells. Other chapters of the book deal with their genotoxic, mutagenic and carcinogenic effects. The toxicity of several metals touch upon the aspects of environmental hazard, ecosystems and human health. Among the cellular responses of heavy metals irregularities in cellular mechanisms such as gene expression, protein folding, stress signaling pathways are among the most important ones. The final chapters deal with biosensors and removal of heavy metals. As everybody is eating, drinking and exposed to heavy metals on a daily basis, the spirit of the book will attract a wide audience.

Book excerpt: Tetraspanins are small (20-50 kDa) integral membrane proteins with four transmembrane domains that have an intrinsic propensity to associate with other membrane proteins and lipids giving rise to the formation of specific tetraspanin-enriched microdomains (TEMs), also referred to as “The tetraspanin web”. In mammals, the tetraspanin family comprises of 33 different members, with the majority of the members being abundantly expressed in almost all cell types, including leukocytes which are responsible for innate and adaptive immunity as well as in other cells that play pivotal roles in immune responses, such as endothelial or stromal cells. Therefore, through the wide range of specific molecular interactions in which they are engaged, tetraspanins influence many processes of up-most relevance in the development, physiology and pathology of the immune system, including the control of immune cell morphology, signaling, adhesion, migration, invasion, fusion, infections and cancer.
